چکیده

1. After incubation of chick-embryo skin slices with [(14)C]proline for 2hr. the specific activities of [(14)C]proline and [(14)C]hydroxyproline in soluble and insoluble collagens and [(14)C]proline in non-collagenous proteins were determined as well as the total amounts of both imino acids in these proteins. On the basis of these results it was demonstrated that soluble collagens having a high proline/hydroxyproline ratio are contaminated with non-collagenous proteins. 2. It was found that, in the presence of a mixture of amino acids in the incubation medium, the rate of synthesis of soluble collagen is significantly decreased. 3. The metabolic activity of collagenous proteins is related to their solubility, but that of non-collagenous proteins is not.

Expression of recombinant human type I-III collagens in the yeast pichia pastoris.

An efficient expression system for recombinant human collagens will have numerous scientific and medical applications. However, most recombinant systems are unsuitable for this purpose, as they do not have sufficient prolyl 4-hydroxylase activity. We have developed methods for producing the three major fibril-forming human collagens, types I, II and III, in the methylotrophic yeast Pichia pasto...

Electron Microscopic and Biochemical Characterization of Collagen in Blattarian Insects

The occurrence of collagen in the cockroach Leucophaea maderae has been demonstrated by electron optical and biochemical techniques. Electron micrographs of tissues of this and a related species (Blaberus craniifer) are presented and they show that collagenous-type fibers occur in the stroma of nonneural as well as neural organs of these insects. Hydroxyproline and hydroxylysine, amino acids co...

Studies on the Biosynthesis of Collagen Ii. the Conversion of 14c-l-proline to 14c-hydroxyproline by Fowl Osteoblasts in Tissue Culture by R. H. Smith, Ph.d.,* and Sylvia Fitton Jackson, Ph.d

The formation of mature mammalian collagen fibre protein requires the incorporation in peptide linkage of, inter alia, 35 residues per cent of glycine, 12 residues per cent of proline, and 10.5 residues per cent of hydroxyproline. According to our present knowledge, in the animal kingdom hydroxyproline occurs only in the collagenous group of proteins and in elastin, which contains 1.5 residues ...

Preparative procedures and purity assessment of collagen proteins.

Collagens represent a large family (25 members identified so far) of closely related proteins. While the preparative procedures for the members that are ubiquitous and present in tissues in large quantities (typically fibre and network forming collagens types I, II, III, IV and V) are well established, the procedures for more recently discovered minor collagen types, namely those possessing lar...
